Microscopic Examination is a rapid screening method for the detection of yeast and bacteria.

Direct microscopic examinations provide qualitative results of the microorganisms and other components. More advanced techniques include qualitative or quantitative viability counts and arrays of digital imaging options.

ETS uses some of the most advanced microscopic techniques available. We perform light microscopy utilizing bright field, phase contrast and Nomarski (differential interference contrast) modes. We also utilize epi-fluorescence to identify specific spoilage microbes.

 Light/phase contrast microscopy is available at all three ETS locations. Standard microscopic results are ready within one working day of sample submission. The St. Helena laboratory has additional microscopic capabilities.

Standard Applications

• Monitor yeast or bacteria starter cultures prior to propagation or transfer
• Scan for suspected spoilage organisms
• Presumptive identification of Brettanomyces based on epi-fluorescence
• Presumptive identification of bacteria based on gram stain reactions
• Estimation of yeast viability using differential staining with methylene blue
• Characterization of sediment or haze as crystalline, organic, or biological in nature
• Total yeast/bacterial counts with haemocytometer
